タイトル: In vivo CRISPR screening directly targeting testicular cells著者: Noguchi, Yuki; Onodera, Yasuhito; Miyamoto, Tatsuo; Maruoka, Masahiro; Kosako, Hidetaka; Suzuki, Jun抄録: CRISPR-Cas9 short guide RNA (sgRNA) library screening is a powerful approach to understand the molecular mechanisms of biological phenomena. However, its in vivo application is currently limited. Here, we developed our previously established in vitro revival screening method into an in vivo one to identify factors involved in spermatogenesis integrity by utilizing sperm capacitation as an indicator. By introducing an sgRNA library into testicular cells, we successfully pinpointed the retinal degeneration 3 (Rd3) gene as a significant factor in spermatogenesis. Single-cell RNA sequencing (scRNA-seq) analysis highlighted the high expression of Rd3 in round spermatids, and proteomics analysis indicated that Rd3 interacts with mitochondria. To search for cell-type-specific signaling pathways based on s ...

タイトル: フェミニスト認識論における無知の問題 --無知の認識論から認識的抑圧へ--著者: 大橋, 一平抄録: The purpose of this paper is to introduce the issue of “epistemology of ignorance, ” which has been discussed as one of the main themes of feminist epistemology, and the process of its formation, while summarizing the state of debate to date and examining how ignorance and credibility are important factors in epistemic injustice and harm. (1) we will outline feminist epistemology and review what this epistemology takes issue with. Then, (2) we will discuss why “ignorance” forms a unique problem domain for the discussion of this epistemology. Based on this, (3) we will organize several perspectives on “pernicious ignorance” as a problem for the epistemology of ignorance. Next, (4) what kind of social structure forms pernicious ignorance will be discussed in relation to credibility judgement. Finally, (5) we will organize the various ...
